Seit 2020 haben wir begonnen, unsere DRFZ-eigenen Formate aufzuzeichen, wie die Hasinger Lecture oder die Vorträge der Avrion-Mitchison-Preisträger. Das werden wir weiter ausbauen.

Long-lasting clonal memory in the innate immune system
Das adaptive Immungedächtnis ist die Grundlage für einen lang anhaltenden Immunschutz gegen Krankheitserreger, kann aber auch rheumatische Erkrankungen aufrechterhalten. Wir konnten zeigen, dass auch das angeborene Immungedächtnis gegen die Infektion mit dem humanen Cytomegalovirus durch klonal expandierte Natürliche Killerzellen stabil aufrechterhalten wird. Diese epigenetische Prägung durch die Stimulation mit aktivierenden Rezeptoren und proinflammatorischen Zytokinen stattet diese Gedächtniszellen mit verbesserten Effektor-Funktionen aus. Der Konzeptnachweis für ein langanhaltendes, klonales Immungedächtnis im angeborenen Immunsystem kann darauf hindeuten, dass dieser Mechanismus nicht nur Immunschutz bietet, sondern auch rheumatische Erkrankungen aufrechterhalten könnte.

ActiMON – Aktivitätsmonitoring bei jungen Rheumatiker*innen
Am DRFZ werden in verschiedenen Forschungsvorhaben Kinder, Jugendliche und junge Erwachsene beobachtet.
Als Teilprojekt des Forschungsverbundes TARISMA wird in ActiMON das Bewegungsverhalten junger Rheumatiker*innen überregional erfasst. Hierbei kommen tragbare Beschleunigungssensoren zum Einsatz, die während einer ganz normalen Woche während aller Wachzeiten getragen werden und die subjektive Einschätzung um objektivierbare Messdaten ergänzen.

Das Deutsche Rheuma-Forschungszentrum Berlin, ein Institut der Leibniz-Gemeinschaft, wurde vor 30 Jahren, 1988, durch das Land Berlin gegründet, dem es für die vielen Jahre der großzügigen Unterstützung und konstruktiven Begleitung zu großem Dank verpflichtet ist. Als einzige außeruniversitäre Forschungseinrichtung in Deutschland zu rheumatischen und muskuloskelettalen Erkrankungen gehört es seit 2009 zur Leibniz-Gemeinschaft und erhält seitdem auch Förderung durch den Bund.
Mit diesem Film werden die Forschungsaufgaben und die Vision des DRFZ anschaulich erklärt.

Das DRFZ untersucht mit grundlagenwissenschaftlichen und epidemiologischen Methoden die Entstehungsbedingungen und Folgen rheumatischer und muskuloskelettaler Erkrankungen. Die biomedizinische Forschung an unserem Institut identifiziert die Zellen, die rheumatische Erkrankungen auslösen und aufrechterhalten. Sie untersucht die zugrunde liegenden molekularen Mechanismen, um Ansätze für ein Eingreifen zu finden. Die epidemiologische Forschung analysiert Risikofaktoren und Langzeitauswirkungen rheumatischer Erkrankungen im Kindes- und Erwachsenenalter und die Angemessenheit der Versorgung. Die Ergebnisse finden Eingang in die klinische Entscheidungsfindung und die Versorgungsplanung.

Interview mit dem Avrion Mitchison-Preisträger 2020, Lennard Ostendorf von der Charité–Universitätsmedizin Berlin, für seine Arbeit zu neuen Erkenntnissen in der Behandlung von Lupus-Patienten.
Das Deutschen Rheuma-Forschungszentrum Berlin (DRFZ), ein Leibniz-Institut vergibt zu Ehren seines Gründungsdirektors Avrion Mitchison jährlich den Avrion Mitchison Preis an junge NachwuchswissenschaftlerInnen, die einen wesentlichen Beitrag zum Verständnis und zur Behandlung von rheumatischen Erkrankungen leisten. Bis 2018 wurde der Preis von der Schering Stiftung finanziert, seit 2019 stiftet ihn das DRFZ. Der Avrion Mitchison Preis ist mit 3.000 Euro dotiert.

The German Rheumatism Research Center Berlin, an institute of the Leibniz Association, was founded 30 years ago, in 1988, by the State of Berlin, to which it is indebted for the many years of generous support and constructive assistance. As the only non-university research institution in Germany on rheumatic and musculoskeletal diseases, it is belonging to the Leibniz Association since 2009 and is also receiving federal funding since then. This film clearly explains the research tasks and the vision of the DRFZ.

By facilitating more than 40-dimensional cytometric analyses on single-cell levels, mass cytometry permits assessing the diversity of leukocytes and other cell types implicated in the pathogenesis or reflecting the pathology of chronic inflammation. The platform also serves for biosignature discovery based on immune cells isolated from patients’ blood samples, facilitating the development of precision medicine in chronic inflammatory diseases. I will discuss the basics, capabilities, limitations and future promises of mass cytometry, and the various techniques that together secure superior quality of mass cytometry data required for successful computational data mining of large data sets, selected data analysis tools, and will highlight applications from my lab.

In female mammals, dosage compensation for X-linked genes is ensured through random X-chromosome inactivation mediated by the long non-coding RNA Xist. During this process most genes are silenced on one randomly chosen X-chromosome. In embryonic stem cells however, where both X-chromosomes are active, double X-dosage affects pluripotency and differentiation through inhibiting the MAPK signaling pathway. We combine theoretical and experimental approaches to understand how X-dosage information will drive cellular decision making processes.

Regina Stark: Regulation of tissue resident memory T cells by transcriptional programs and their microenvironment

Tissue resident memory T cells (TRM) are non-circulating immune cells that contribute to the first line of local defense against reinfections. Especially for infection of mucosal surfaces they confer superior protective capacities over circulating memory T cells. We recently discovered transcriptional networks that guide the generation and maintenance of human and murine TRM. These data demonstrate that TRM are not simply memory T cells at an unexpected location but cells with a transcriptional program distinct from circulating memory T cells. Next to the transcriptional landscape we also analyzed the influence of the microenvironment on TRM regulation. The location of TRM at hot-spots of pathogen encounter frequently exposes them to tissue damage and inflammation. This history of danger signal exposure is an important aspect of TRM mediated immunity that has been overlooked so far. We discovered a novel pathway of sensing of tissue damage selectively employed by TRM but not circulating T cells, which demonstrates that tonic survival signals as well as inflammatory events shape local immune protection.

We are investigating how the immune system protects the host against lung infections and how those defense mechanisms re influenced by endogenous and exogenous susceptibility factors. I will present our latest insight and discuss how a better understanding of these mechanisms might help to develop novel prophylactic and/or therapeutic strategies.

Summary: The airways and the lung are constantly exposed to microbial and environmental stimuli. As the lung’s main physiological function of gas exchange critically relies on its delicate microarchitecture, complex regulatory circuits have evolved to balance inflammatory immune responses. Yet, the prevalence of inflammatory disorders such as asthma is increasing, probably due to altered life styles and environments. I will discuss new insights into the pathophysiology, classification, and treatments f airway diseases. I will also discuss new data from our laboratory on the role of regulatory cells during bacterial superinfections in the lung.

Circulatory and tissue-resident lymphocytes are key players of the skin immune system and are involved in both protective and pathologic skin immune responses. We analyzed the role of T cell tissue egress in T cell mediated skin inflammation as well as the potential of IL-10 producing innate-like B cells to migrate into skin. The results demonstrate novel potential pathways in the regulation of skin inflammation.

Chronische Entzündung kann durch alleinige Fehlregulation im angeborenen Immunsystem (Autoinflammation) oder durch einen Zusammenbruch der Selbst-Toleranz im T- und B-Zellsystem (Autoimmunität) verursacht werden. Autoimmunität ist meist Folge eines komplexen Zusammenwirkens von angeborener und adaptiver Immunität. Unkontrollierte antivirale Typ I-Interferonantworten des angeborenen Immunsystems sind typisches Kennzeichen der polygenen, multifaktoriellen Autoimmunkrankheit SLE. Seltene monogene Varianten des SLE erleichtern die Aufklärung von Mechanismen, welche die pathogene Interferonproduktion induzieren, z.B. die Aktivierung zytosolischer Nukleinsäuresensoren durch intrazelluläre Akkumulation von endogenem
Nukleinsäure-Abfall. Dies illustriert eindrücklich die Auslösung einer Autoimmunerkrankung durch chronische Aktivierung des angeborenen Immunsystems.

Die Ursachen für chronisch entzündliche Erkrankungen, wie die rheumatoide Arthritis, die Schuppenflechte (Psoriasis) und die Multiple Sklerose sind unklar. Es gibt Hinweise darauf, dass entartete Immunzellen und Autoimmunität diese Krankheiten hervorbringt. Professor Becher wird die Grundlagen für diese Hypothese kurz darstellen. Damit das Immunsystem gut funktioniert und uns gegen Krankheitserreger wie Viren und Bakterien schützen kann, muss die Kommunikation zwischen Immunzellen (weissen Blutzellen) gut funktionieren. Professor Becher wird anhand von jüngsten Forschungsergebnissen Hinweise dafür bringen, dass die chronischen Entzündungserkrankungen vor allem eine Misskommunikation unter Immunzellen zu Grunde liegt.

Was leistet die Grundlagenforschung im Bereich chronischer Infektionen?

Chronische Infektionen stellen weltweit ein hochaktuelles und wissenschaftlich nach wie vor weitgehend ungelöstes medizinisches Problem dar. Zu erwähnen sind in diesem Zusammenhang die mehr als 30 Millionen HIV Infizierten als auch die geschätzten 300-600 Millionen Patienten mit chronischen Hepatitis B oder C Infektionen. Im Gegensatz zu chronischen HBV Infektionen wurden erfreulicherweise in den letzten Jahren wesentliche Fortschritte in der Behandlung von chronischen HCV Erkrankungen und bei der Kontrolle bestehender HIV Infektionen erreicht. Trotzdem ist die Mehrheit der Patienten noch immer von diesen Therapien aus verschiedenen, meist ökonomischen Gründen ausgeschlossen. Diese und viele weitere nicht genannte Beispiele unterstreichen die Wichtigkeit der Erforschung von Immunantworten in chronischen Infektionen, um dadurch die Entwicklung neuer und vor allem kosteneffizienter Therapien zu stimulieren.

In der Grundlagenforschung stellt sich vor diesem Hintergrund vor allem die Frage, warum jene Abwehrmechanismen, die uns beispielweise gegen eine echte Grippe oder eine Hepatitis A Virusinfektion effektiv schützen, im Falle der zuvor genannten Infektionen in der Regel versagen. Gleichbedeutend ist auch die Frage, wie Immunantworten so beeinflusst oder manipuliert werden können, um vor oder auch nach einer Infektion mit einem problematischen Erreger einen ausreichenden Schutz zu erreichen oder sicher zu stellen. Ziel des Vortrags ist es, Einblicke in den Stand der Forschung zu diesem Thema zu gewähren und grundlegende Prinzipen zu illustrieren, wie sich Immunantworten in kontrollierten und in aktiv-persistierenden Infektionen unterscheiden. Weitere Schwerpunkte sind die Darstellung neuester Ergebnisse und deren therapeutischer Implikationen sowie Verweise, wie die Grundlagenforschung im Bereich chronischer Infektionen bahnbrechende Erkenntnisse zur Therapie von malignen Erkrankungen geleistet hat.
